Abstract
Mesothelin is shed from cells by the action of membrane bound proteases. We have identified the shedding sites using mesothelin shed into ascites of cancer patients. We have prepared a monoclonal antibody 15B6 that binds at the shedding sites and inhibits mesothelin shedding by cell lines. We believe shedding is important for the carcinogenic effect of mesothelin. We plan to test them to determine if the antibody affects spread of mesothelin expressing tumors in mice. We will use tumors that grow in the peritoneal cavity and tumors that grow subcut to initiate these studies.
