Characterizing molecular interactomes using TurboID

Abstract

During this period, the collaborative team has developed tools to explore new uses of TurboID as a proximity labeling tool to define small molecule interactomes. A variety of TurboID constructs and expression strategies were explored to characterize the behavior of the system. Several proof-of-concept small molecules, with known mechanisms of action, including dasatinib and methotrexate have been employed for these studies. The team has also pursued an optimized method for isolating biotinylated proteins for subsequent mass spectrometry and target identification.
