iPSCs to study Diversity in Alzheimers disease and Alzheimers disease-related dementias (iDA)

Abstract
We have coordinated the shipment of 200 PBMC samples from genetically diverse disease and control individuals from the ADNI/National Centralized Repository for Alzheimers Disease (NCRAD) to the New York Stem Cell Foundation (NYSCF) to generate iPSC lines. NYSCF began processing the samples in September 2022, with the first iPSC lines finished May 2023. Lines will continue to be generated until at least February 2024 and shipped to NCRAD on a rolling basis until all 200 are complete. As lines are received by NCRAD, CARD will begin characterization experiments in differentiated neurons, astrocytes, and microglia. CARD will also produce SNP array and long-read sequencing data of iPSCs that will be publicly available through the Alzheimers Disease Workbench (ADWB). CARD has also established a collaboration with industry partner Amprion, which will use their proprietary seed amplification assay (SAA) to detect aggregates of misfolded -synuclein in CSF samples from ADNI. Amprion is currently processing an initial batch of 144 samples, with additional samples planned for FY24.
