Predicting Chromatin Signatures of Alzheimers Disease and Related Dementia from Peripheral Blood

Abstract

In the past year, we have performed pilot experiments to determine the feasibility of this project. We generated single-nucleus ATAC, single-cell RNA and single-nucleus RNA sequencing (snATAC-seq, scRNA-seq and snRNA-seq respectively) libraries from multiple peripheral blood mononuclear cell (PBMC) samples. We have optimized and determined various steps and thresholds in the process including minimum required cell number, minimum required viability, enrichment protocols, dead cell removal, bead and antibody titrations etc. Furthermore, we performed immunophenotyping to identify cell population changes in an orthogonal technique. We are currently engaged in analyzing our pilot data and establishing computational methods to further analyze the data.
