Abstract
Project Summary/Abstract We have recently developed a microscopy-based method that, for the first time, enables interrogation of histone modifications at single loci in single cells. This method is termed SCEPTRE (for Single-Cell Evaluation of Post TRanslational Epigenetic encoding) and utilizes super-resolution imaging to directly resolve and quantify histone modifications at specific DNA or gene loci that are labeled by immunofluorescence and DNA fluorescence in situ hybridization (FISH), respectively. We propose to develop SCEPTRE, a method currently in use with cultured or isolated cells, for epigenetic profiling of kidney tissues and to use SCEPTRE to study healthy aging and senescence in mouse kidney tissues.
