Abstract

Project Summary This project is an administrative supplement request in response to NOT-GM-22-017. The objective of this project is to expand the capabilities of the CRISPR-IMAGE research support core as part of parent award (COBRE, 5P20GM135008) with a microscope to enable optogenetic manipulating and imaging of signaling dynamics, molecular reorganization, and cellular movements on the time scale of inflammation and resolution (hours to days). This instrumentation’s implementation capitalizes on new expertise brought into the COBRE center with the hire of a new director for our CRIPSR-IMAGE core. This direction will expand our institutional capacity for the analyses of cellular-to-tissue inflammatory process by direct visualization and quantification of cellular dynamics during cellular and tissue-level processes such as differentiation, immune polarization, and responses to oxidative stress. In addition to increasing our capacity for research within the COBRE, this instrument will be used for graduate education via a hands-on course taught by the director (with currently 18 students/year) and it will be available to users from across campus studying diverse cellular processes ranging from bacterial to plant systems.
