Abstract

Summary The PI is requesting an administrative supplement to fund the purchase of an isothermal titration calorimeter (ITC), specifically a Malvern MicroCal PEAQ ITC. The instrument will be used as a label-free approach to measure binding constants (Kd or Ka) and thermodynamics (ΔH and ΔS) of enzyme-ligand interactions key to experimental questions described in the parent award (1R35GM147276). This instrument will enable us to perform more types of enzyme-ligand experiments than possible with our current equipment and to perform the previously proposed experiments with greater confidence. All experiments are still in line with the original scope of the parent award.
