Abstract
We have reconstituted lipid droplet biogenesis in vitro. It requires no proteins other than a triglyceride synthesizing protein. We are using this system to understand how lipid compositon and biophysical properties of membranes affect biogenesis. Longer-term, we will add proteins know to play a role in lipid droplet biogenesis to determine how they function. We have undertaken a large-scale RNAi screen in collaboration with NCATS to identify genes required for VLL biogenesis and secretion from hepatocytes. We are performing a CRISPRi screen to identify genes required for peroxisome biogenesis. This screen is being gone in collaboration with Richard Youles lab. It will be the first open-ended screen for peroxisome biogenesis factor in mammalian cells.
