Abstract

COVID 19 related projects: Humanized animals: Using CRISPR/cas9 mediated gene KI via specific gRNAs and ODNTs, several lines have been created with the human ACE2 mutations individually in exons #2, exon #3 and exon #9. In the second stage, the sperm from these lines was used to create embryos that were targeted for second exon and created lines with dual mutations in exon#2 and exon#9 which will be used to create triple mutants humanized animals. COVID-funding: MGGM was awarded additional $36,000 for the creation of humanized ACE2 NOD/ShiLtJ mice as model for underlying conditions in humans. Revised proposal was submitted for humanizing ACE2 gene of the Thicket rat as a test model in collaboration with Dr. Patrick Duffy and Dr. Heather Hickman. Novel ES cell lines created from the Thicket rat will be utilized for creating humanized animals via tetraploid mouse embryo complementation as placenta donor
