Structure-Function Studies Of DNA Replication Fidelity

Abstract

This year, our group published five scientific articles related to DNA replication and its fidelity. One described how equal replication fidelity is achieved on both DNA strands, especially including via the process of extrinsic proofreading by the lagging strand DNA polymerase delta. Another is a review of the incorporation of ribonucleotide triphosphates into DNA during replication, how they are repaired, and the consequences of repair. A third article describes the X-ray crystal structure and the processing of topoisomerase 1-triggered DNA damage by Apn2 nuclease. The fourth publication describes the mutagenic consequences of failure to accurately replicate nuclear DNA by mutant derivatives of the leading strand replicase with amino acids substitutions associated with cancer in humans. The fifth article reviews to contribution of extrinsic proofreading by DNA polymerase delta to replication fidelity.
