Abstract
HA22 is licensed to Medimmune and been given the name Moxetumomab pasudotox (Moxe). It has been tested in patients with advanced chemo-resistant hairy cell leukemia (HCL) in adults and in acute lymphoblastic leukemia in children. The HCL phase 3 trial has met its FDA mandated efficacy endpoint and the agent and was approved by the FDA in 2018. New trials with Moxe are being conducted with Rituxan to help improve the complete response rate and to eliminate MRD. 12 patients have been treated and the results look encouraging. LMB-100 is an immunotoxin targeting mesothelin expressing cancers. In collaboration with Drs Hassan and Alewine, we treated patients intravenously but saw very little activity. Because laboratory studies in mice showed that intra-tumoral injection of LMB-100 caused tumor regressions and anti-tumor immunity, we have recently opened a protocol where LMB-100 is injected directly into tumors of mesothelioma patients by an interventional radiologist and the patients also receives Ipilimumab IV. The first patient was recently treated. We are also collaborating with Darell Bigner at Duke who treats brain tumors with immunotoxins and with Jay Berzofsky in CCR who has developed a vaccine to treat prostate cancer that targets the TARP protein discovered in LMB.
